    Get PDF
The aim of the MoreGrasp project is to develop a noninvasive, multimodal user interface including a brain-computer interface (BCI) for intuitive control of a grasp neuroprosthesis to support individuals with high spinal cord injury (SCI) in everyday activities. We describe the current state of the project, including the EEG system, preliminary results of natural movements decoding in people with SCI, the new electrode concept for the grasp neuroprosthesis, the shared control architecture behind the system and the implementation of a user-centered design

The use of high-throughput sequencing to investigate an outbreak of glycopeptide-resistant Enterococcus faecium with a novel quinupristin-dalfopristin resistance mechanism.

High-throughput sequencing (HTS) has successfully identified novel resistance genes in enterococci and determined clonal relatedness in outbreak analysis. We report the use of HTS to investigate two concurrent outbreaks of glycopeptide-resistant Enterococcus faecium (GRE) with an uncharacterised resistance mechanism to quinupristin-dalfopristin (QD).Seven QD-resistant and five QD-susceptible GRE isolates from a two-centre outbreak were studied. HTS was performed to identify genes or predicted proteins that were associated with the QD-resistant phenotype. MLST and SNP typing on HTS data was used to determine clonal relatedness.Comparative genomic analysis confirmed this GRE outbreak involved two distinct clones (ST80 and ST192). HTS confirmed the absence of known QD resistance genes, suggesting a novel mechanism was conferring resistance. Genomic analysis identified two significant genetic determinants with explanatory power for the high level of QD resistance in the ST80 QD-resistant clone: an additional 56aa leader sequence at the N-terminus of the lsaE gene and a transposon containing seven genes encoding proteins with possible drug or drug-target modification activities. However, HTS was unable to conclusively determine the QD resistance mechanism and did not reveal any genetic basis for QD resistance in the ST192 clone. This study highlights the usefulness of HTS in deciphering the degree of relatedness in two concurrent GRE outbreaks. Although HTS was able to reveal some genetic candidates for uncharacterised QD resistance, this study demonstrates the limitations of HTS as a tool for identifying putative determinants of resistance to QD

A Moving Boundary Flux Stabilization Method for Cartesian Cut-Cell Grids using Directional Operator Splitting

An explicit moving boundary method for the numerical solution of time-dependent hyperbolic conservation laws on grids produced by the intersection of complex geometries with a regular Cartesian grid is presented. As it employs directional operator splitting, implementation of the scheme is rather straightforward. Extending the method for static walls from Klein et al., Phil. Trans. Roy. Soc., A367, no. 1907, 4559-4575 (2009), the scheme calculates fluxes needed for a conservative update of the near-wall cut-cells as linear combinations of standard fluxes from a one-dimensional extended stencil. Here the standard fluxes are those obtained without regard to the small sub-cell problem, and the linear combination weights involve detailed information regarding the cut-cell geometry. This linear combination of standard fluxes stabilizes the updates such that the time-step yielding marginal stability for arbitrarily small cut-cells is of the same order as that for regular cells. Moreover, it renders the approach compatible with a wide range of existing numerical flux-approximation methods. The scheme is extended here to time dependent rigid boundaries by reformulating the linear combination weights of the stabilizing flux stencil to account for the time dependence of cut-cell volume and interface area fractions. The two-dimensional tests discussed include advection in a channel oriented at an oblique angle to the Cartesian computational mesh, cylinders with circular and triangular cross-section passing through a stationary shock wave, a piston moving through an open-ended shock tube, and the flow around an oscillating NACA 0012 aerofoil profile.Comment: 30 pages, 27 figures, 3 table

Can landscape-scale approaches to conservation management resolve biodiversity-ecosystem service trade-offs?

Conservation management is increasingly being required to support both the provision of ecosystem services and maintenance of biodiversity. However, trade-offs can occur between biodiversity and ecosystems services. We examine whether such trade-offs can be resolved through landscape-scale approaches to management. We analysed the biodiversity value and provision of selected ecosystem services (carbon storage, recreation, aesthetic and timber value) on patches of lowland heathland in the southern English county of Dorset. We used transition matrices of vegetation dynamics across 112 heathland patches to forecast biodiversity and ecosystem service provision on patches of different sizes over a 27-year timeline. Management scenarios simulated the removal of scrub and woodland and compared (i) no management (NM); (ii) all heaths managed equally (AM); and management focused on (iii) small heaths (SM) and (iv) large heaths (LM). Results highlighted a number of trade-offs. Whereas biodiversity values were significantly lower in woodland than in dry and humid heath, timber, carbon storage and aesthetic values were highest in woodland. While recreation value was positively related to dry heath area, it was negatively related to woodland area. Multicriteria analysis ranked NM highest for aesthetic value, carbon storage and timber value. In contrast, SM ranked highest for recreation and LM highest for biodiversity value. In no scenario did the current site-based approach to management (AM) rank highest. Synthesis and applications. Biodiversity-ecosystem service trade-offs are reported in lowland heathland, an ecosystem type of high conservation value. Trade-offs can be addressed through a landscape-scale approach to management, by varying interventions according to heathland patch size. Specifically, if management for biodiversity conservation is focused on larger patches, the aesthetic, carbon storage and timber value of smaller patches would increase, as a result of woody succession. In this way, individual heathland patches of either relatively high biodiversity value or high value for provision of ecosystem services could both potentially be delivered at the landscape scale
